Helixmith is a research-oriented venture company established with the aim of developing innovative biopharmaceuticals, especially as the First Mover in the field of gene therapy for cardiovascular and neuro degenerative diseases. It started as a spin-off venture at Seoul National University in 1996 and has been listed on the KOSDAQ since 2006. The number of employees, which was only three when established, currently reaches 140 in Korea and the United States altogether, including 60 researchers. Helixmith aims to treat incurable diseases with high prevalence rate. The most advanced flagship product is Engensis, a novel plasmid DNA. Engensis is engineered to express both wild type isoforms of HGF based on its mechanism of inducing angiogenesis and neuronal regeneration. It is currently on its phase 3 clinical trials for diabetic peripheral neuropathy and intractable foot ulcers in the United States and phase 2 clinical trials are underway for Lou Gehrig's disease and claudication. In addition to Engensis, various types of innovative bio-new drugs based on AAV, antibody, and CAR-T technology are under development. In order to produce gene therapy products and enter rapidly growing gene therapy CDMO business, Helixmith recently completed its GMP facility construction. Instruction | Synthetic biology has emerged as a new approach for re-designing biological systems, and even engineering new ones that are optimized for specific medical, industrial, and biotechnological applications. It is expected that the various disciplines of synthetic biology will soon bring out disruptive technology solutions to open up the era of Bioeconomy. On the other hand, however, the complicated and unpredictable nature of living systems often limits the reach and scope of these efforts. Cell-free systems, which reproduces biological synthetic reactions in programmable liquid, can address many of the issues associated with the cell-based approaches. This session was organized to discuss the present and future of cell-free synthetic biology. Instruction | Artificial cells, cell engineering and therapy are emerging technologies which will make a significant impact on the future of medicine and healthcare. Cell engineering is a field that has progressed steadily over the last few decades, currently reaching a level of development that enables practical applications in human patients. Cellular therapies are designed to improve the immune system's ability to fight cancer and disease. There are several approaches for altering the cellular processes such as silencing or overexpressing of targeted genes and altering expression of genes using microRNA (miRNA). These cell therapies offer the promise of treating and altering the course of diseases which cannot be addressed adequately by existing pharmaceuticals. | ||

Instruction | Recently technologies for neutralization of carbon 2050 are urgently demanded. Aromatics originated from lignin will be valuable as well as carbon neutral, however, it has been known very hard to break down lignin into valuable aromatic compounds owing to chemical recalcitrance. Biorefinery based on solely sugars is facing up difficulties to meet the technol-economic feasibility, which strongly implies that lignin valorization will be key issue for the success of biorefinery process. For this symposium, currently emerging technology will be introduced for vitalization of lignin biorefinery. Converging technology between chemical and biotechnology will be crucial for the development of enabling technology to valorize lignin effectively. From Lab. Research to Plant Design: Bioprocess Modeling and Analysis |

Instruction | Success in the bioindustry relies on breakthrough technology, which spends vast time and money in the R&D stage, and the implementation and commercialization of that technology also require considerable investment. For decision-making before the investment, the process modeling, optimal arrangement of units, and analysis/optimization should be performed based on a comprehensive understanding of the process developed. In particular, as the white biotech processes have recently been in the spotlight as a promising option for carbon-neutrality, the scale-up and commercialization activity are becoming significant, and thus TEA (Techno-Economic Analysis) and LCA (Life Cycle Analysis) for the bioprocess is indispensable. This symposium presents the state-of-the-art results of process design and analysis for R&DD. The technical and environmental features of bioprocesses for the construction of "Net-Zero Society" in the future will be discussed through the case study in other research fields together. | ||

The current manufacturing processes for viral vectors for gene therapy, which we can define as Gene Therapy Manufacturing 1.0 (e.g., adherent cell culture and transient expression from plasmid transfection), are not productive enough to meet the future demand considering the quickly increasing number of approved gene therapies and clinical trials. A transition is therefore ongoing to implement more productive and scalable processes, leading to Gene Therapy Manufacturing 2.0 using suspension cell culture and developing producer cell lines. Novel strategies towards enhancing enzymatic properties such as catalytic activity, specificity and stability have opened up a plethora of novel applications for industrial production, agriculture and sustainability. Such improved enzymes are typically identified in high throughput functional screening campaigns by either investigating the vast number of naturally occurring microbial homologs or by exploring the synthetic combinatorial space of given amino acid motifs. Both strategies rely on the ability to generate high numbers of DNA templates. At Twist Biosciences we have revolutionized DNA synthesis in terms of scale, uniformity and error rate. The resulting high-quality products such as clonal genes and DNA variant libraries comprise the ideal basis for high throughput functional screenings for enhanced enzyme variants. For the synthetic biologist, biological building blocks are often generated from bacteria. The design-build-test process is utilized in order to engineer these building blocks. This often requires screening and picking colonies of interest. Traditionally, colony picking is performed manually using sterile pipette tips, toothpicks, or inoculation loops, which is a slow, labor-intensive, and time consuming process. Synthetic biology research can, therefore, benefit greatly from the flexibility and throughput of automated colony pickers. The versatility of fully automated bacterial colony picker in synthetic biology | |
기존 이차원 단층세포 배양 기술은, 실험적 편의성과 값싼 비용이라는 장점을 바탕으로 누구나 쉽게 체외에서 특정세포를 배양할 수 있는 연구플랫폼으로 자리잡았으며, 이렇게 배양된 단층배양세포는 개발중인 신약의 성능과 독성을 평가하기위한 보편적인 in vitro 모델로 널리 사용되어 왔습니다. 그러나, 현저하게 낮은 단층배양 세포의 생체 모사성 때문에 이를 기반으로 도출된 연구결과가 실제 임상에서 재현되지 못하는 사례가 빈번하고, 실제로 전임상 및 임상연구의 높은 실패율을 야기하는 치명적인 한계를 보여주고 있습니다.
