Prof. Shuichi Takayama Georgia Institute of Technology USA H-index=81 Organoid, Cellchip, Organic chemistry Professor, GRA Eminent Scholar Price Gilbert, Jr. Chair in Regenerative Engineering and Medicine	Background Dr. Shu Takayama earned his BS and MS in Agricultural Chemistry at the University of Tokyo. He earned a PhD in Chemistry at The Scripps Research Institute in La Jolla, California studying bio-organic synthesis with Dr. Chi‐Huey Wong. He then worked as a postdoc with Dr. George Whitesides at Harvard University where he focused on applying microfluidics to studying cell and molecular biology. Dr. Takayama began his career at the University of Michigan, where led his lab in the Department of Biomedical Engineering and Macromolecular Science & Engineering for over 17 years. In 2017, the lab moved to Georgia Tech where Shu became the Georgia Research Alliance Price Gilbert Chair Professor of Biomedical Engineering in the Wallace H. Coulter Department of Biomedical Engineering. Dr. Takayama’s research interests are diverse and motivated by clinical and biotechnology needs. He is always interested in hearing from stakeholders in these areas who are seeking engineering collaboration.

Background

Dr. Shu Takayama earned his BS and MS in Agricultural Chemistry at the University of Tokyo. He earned a PhD in Chemistry at The Scripps Research Institute in La Jolla, California studying bio-organic synthesis with Dr. Chi‐Huey Wong. He then worked as a postdoc with Dr. George Whitesides at Harvard University where he focused on applying microfluidics to studying cell and molecular biology.
Dr. Takayama began his career at the University of Michigan, where led his lab in the Department of Biomedical Engineering and Macromolecular Science & Engineering for over 17 years. In 2017, the lab moved to Georgia Tech where Shu became the Georgia Research Alliance Price Gilbert Chair Professor of Biomedical Engineering in the Wallace H. Coulter Department of Biomedical Engineering.
Dr. Takayama’s research interests are diverse and motivated by clinical and biotechnology needs. He is always interested in hearing from stakeholders in these areas who are seeking engineering collaboration.

Systems Biology of Yeast Metabolism

Metabolic Engineering relies on a thorough understanding of how the many different metabolic reactions in the cell to be engineered interacts. Genome-scale metabolic models offers a very strong tool for performing quantitative analysis of how the many different reactions in the metabolic network interacts, and through the addition of kinetic and proteome constraints to these models their predictive strength has significantly improved. However, these models can also be used for integrative analysis of quantitative data, e.g. proteomics and metabolomics data. In the lecture there will be presented progress on how kinetic and proteome constraints can improve the predictive strength of genome-scale metabolic models for use in metabolic engineering. Examples will be given of both identification of novel metabolic engineering designs and of using these models for gaining novel insight into the functioning of metabolism.

With his lab members he brings the tools of systems biology to bear on the challenges of sustainability. His research aims to understand the cellular highways of energy and carbon transformations in quantitative terms both within living cells and in the global environment. He is passionate about better understanding the world with the aid of numbers and is the author of “Cell biology by the numbers”, translated to six languages. His lab focuses on the following research topics: Enhancing carbon fixation. Combining synthetic biology with metabolic modeling we design and implement novel carbon fixation pathways in E. coli, that can convert atmospheric CO₂ into food, fuels and fine chemicals. Quantifying the anthropocene. Performance of a global census of the biosphere biomass with a focus on wild-mammals, arthropods and plants, enabling us to portray the effects of humanity. Autotrophic growth was achieved following several months of continuous laboratory evolution in a chemostat under intensifying organic carbon limitation and confirmed via isotopic labeling.

He is a member of the National Academy of Engineering and the National Academy of Inventors Presentation Engineered Polyketide Synthases as Platform for Synthetic Chemistry Polyketides are one of the largest classes of natural products, possessing immense structural diversity and complex chemical architectures. Many polyketides (PKs) are among the most important secondary metabolites for their applications in medicine, agriculture, and industry. Examples include anticancer drugs (epothilone), antibiotics (erythromycin), insecticides (spinosyn A) and antifungals (amphotericin B). These particular examples of polyketides are biosynthesized by multimodular enzyme complexes known as type I modular polyketide synthases (PKSs). Working in an assembly-line fashion, multimodular PKSs assemble and tailor readily available acylCoAs within the host cell into large, complex, chiral molecules. Each of these PKSs comprises a series of modules that can be further dissected into a series of domains responsible for the extension of the polyketide back- bone through condensation and selective reductive processing of an acyl-CoA building block. The collinear architecture of these modules, apparent by inspection of the domains present and the predictive selectivity motifs harbored within, provide insights into the chemical connectivity and stereochemical configuration of the polyketide metabolite from analysis of its coding sequence. While PKSs have been traditionally studied for the production of pharmaceuticals, engineered modular PKSs have the potential to be an extraordinarily effective retrosynthesis platform for the bio-production of products from biofuels and commodity chemicals to both pharmaceutical and nonpharmaceutical fine and specialty chemicals. By rearranging existing polyketide modules and domains, one can exquisitely control chemical structure from DNA sequence alone. However, this potential has only just begun to be realized as the compounds that have been made using engineered PKSs represent a small fraction of the potentially accessible chemical space. In my talk, I will highlight work from our laboratory where we have engineered PKSs to produce a variety of commodity and specialty chemicals and developed software and high throughput robotic platforms to enable design and construction of PKSs in high throughput.

Engineered Polyketide Synthases as Platform for Synthetic Chemistry

Polyketides are one of the largest classes of natural products, possessing immense structural diversity and complex chemical architectures. Many polyketides (PKs) are among the most important secondary metabolites for their applications in medicine, agriculture, and industry. Examples include anticancer drugs (epothilone), antibiotics (erythromycin), insecticides (spinosyn A) and antifungals (amphotericin B). These particular examples of polyketides are biosynthesized by multimodular enzyme complexes known as type I modular polyketide synthases (PKSs). These make us more difficult to classify and standardize odors. More than one hudred years ago, Alexander Graham Bell, who invented the telephone, said that we have very many different kinds of smells, but until you can measure their likenesses and differences you can have no science of odor. However, it is still difficult to measure odors. Why is it so difficult? Because we have not understood the identity of 'odor' until Robert Axel and Linda Buck discover the mechanism of smell sensing on the molecular basis. Now we know that information of an odor is the combinatorial pattern made of the olfactory receptors activated by the odor molecules. Choul-Gyun Lee Department of Biological Sciences and Bioengineering, Inha University, Incheon, Korea	Blue Biotechnology: A Feasible Way to a Sustainable Future Conventional biomass has a limited sustainability and could negatively affect the environment and food security. Replacing chemicals from fossil fuels requires a significant quantity of biomass and an enormous area due to the low photosynthetic efficiency. Furthermore, water scarcity and peak phosphorus are also of concerns for mass production of biomass feedstocks for biorefinery and white biotechnology. One of the viable options to resolve this situation is blue biomass or marine photosynthetic organisms culturing with seawater. Various types of algal culture systems have been developed to improve sustainability and economic feasibility of blue biomass production. We have developed various floating microalgal culture systems using Selectively-Permeable Materials (SPMs) so that the microalgae can grow on dissolved eutrophic compounds in seawater. We’ve found that the ion permeability of SPMs is one of the key limiting factors and increasing ion permeability resulted in enhanced blue biomass productivity. In other words, our ocean culture systems are not light limited, yet, though we have achieved average biomass productivity of 20 g/m2/day using just seawater on our ocean test-beds. Blue Biotechnology: A Feasible Way to a Sustainable Future

Conventional biomass has a limited sustainability and could negatively affect the environment and food security. Replacing chemicals from fossil fuels requires a significant quantity of biomass and an enormous area due to the low photosynthetic efficiency. Furthermore, water scarcity and peak phosphorus are also of concerns for mass production of biomass feedstocks for biorefinery and white biotechnology. One of the viable options to resolve this situation is blue biomass or marine photosynthetic organisms culturing with seawater. Various types of algal culture systems have been developed to improve sustainability and economic feasibility of blue biomass production.

We have developed various floating microalgal culture systems using Selectively-Permeable Materials (SPMs) so that the microalgae can grow on dissolved eutrophic compounds in seawater. We’ve found that the ion permeability of SPMs is one of the key limiting factors and increasing ion permeability resulted in enhanced blue biomass productivity. In other words, our ocean culture systems are not light limited, yet, though we have achieved average biomass productivity of 20 g/m2/day using just seawater on our ocean test-beds. The feasible direction of blue biomass for sustainable future feedstock for biorefinery will be discussed.

기술 세부 내용

○ 시장에서 판매 중인 2,3-부탄다이올은 화학촉매공정을 통해 이성질체 혼합물 (racemic mixture)로 생산되며, 특정 이성질체로 분리할 경우 공정비용 증가로 그 가격이 매우 고가임
(2,3-부탄다이올은 meso, levo, dextro 3가지 이성질체가 존재함)

○ GS칼텍스는 독자적인 미생물개량 및 발효기술 개발을 통해 meso나 levo 이성질체를 고선택도 고수율로 2,3-부탄다이올을 생산할 수 있는 기술을 확보하였음

○ 자체 개발한 분리정제 및 탈색탈취 기술을 통해 무색, 무취의 고순도 2,3-부탄다이올 회수기술을 확보하였음

○ 유전자조작을 하지 않은(Non-GMO) 바이오매스 원료 및 미생물을 사용하고, 분리 정제 과정에서도 끓는점, 크기 등 물리적인 특징만을 이용해 친환경적으로 생산하고 있음

○ 2,3-부탄다이올은 식물, 동물, 토양 등 자연에 널리 존재하는 천연물질이지만, 케미칼 시장에서는 널리 사용되고 있지 않는 신물질로 GS칼텍스가 바이오화학기술을 통해 대량생산에 성공하였으며,
기술개발과 함께 자체 용도 개발을 통해 세계에서 유일하게 화장품 원료 등으로 시장에 진입하여 판매 확대 중에 있음

기술 세부 내용

세포 배양의 기술은 싱글유즈 (Single-use) 장비를 이용한 바이오리액터 활용 및 업스트림 공정 개발 전략을 다룹니다. 공정 최적화 및 스케일-업을 위한 중요 변수 전반에 대한 것입니다. ReadyToProcess WAVE는 single-use bag 내부의 용액 움직임에 의한 믹싱 및 기체 전달 기능을 극대화한 single-use rocking bioreactor입니다. 세포 건강에 악영향을 주는 shear stress가 적고 single-use bag 사용으로 교차 오염 위험이 없어 연구 단계나 GMP 초기 종배양에 적합한 기술입니다.

단백질 분리 정제 (Protein purification)를 위한 기술과 방법의 개발은 생물공학분야에서 많은 발전을 위한 필수적인 전제조건이 되어져 왔다. 단백질 분리 정제는 간단한 one-step purification 과정에서부터 증명된 대량 정제의 생산 과정에까지 다양하다. 원하는 purity를 얻기 위해서는 종종 한번의 정제 과정 이상이 필요로 되어진다. 성공적인 단백질 정제는 적절한 technique을 선택하고, 그 방법으로 최상의 결과가 나오도록 optimization 하고, 수율 (yield)은 최대로 얻고 요구되는 정제 단계를 최소로 하는 것이다. 대부분의 단백질 정제는 크로마토그래피 (chromatography) 방법을 포함한다. 그 결과로 크로마토그래피는 단백질 정제를 위한 가장 필수적인 기술이다.

In particular, huHF has a specific and strong affinity for transferrin receptors on dendritic cells, suggesting that optimal integration of tumorꠓspecific antigens (TSA) into huHF and administration of the TSA-integrated huHF guide efficient activation of tumor-specific anti-tumor immunity in the body. Further, huHF platform enables high-capacity loading and cancer cell-specific activation of various chemotherapeutic drugs such as doxorubicin, mitomycin C, and gemcitabine that are still clinically useful for cancer treatment. Despite considerable therapeutic efficacy, severe toxicity-associated problems of the chemotherapeutic drugs, which are mainly caused by the non-specific mode of action such as chromosomal DNA damage and/or interference in the DNA replication even in normal cells, remain unresolved. Here I introduce recent important results of my research team in the huHF platformꠓbased cancer immuno- and chemotherapy, showing that both drug toxicity and efficacy in in vivo cancer treatment were dramatically enhanced. This approach is expected to open up a novel clinical route to break through the limits of and thus bring about safer and more widespread adoption of current cancer therapy.

Recent Trends in the Development of Therapeutic Biologics

Designing and engineering new biologics has played essential roles in developing novel therapeutics and expanding the scope of medicine. This session covers representative works of developing next-generation therapeutic biologics against various diseases. The first speaker (Prof. Kim) presents an innovative approach to using endogenous virus-specific CD8+ T cells for cancers based on tumor-specific cytosol-penetrating antibodies. Prof. Jung describes various antibody Fc variants his group has developed so far, which can endow superior functions to antibody therapeutics. Then, Prof. Van Deventer presents a new platform technology to discover crosslinkable antibodies and its application in developing inhibitors against a Botulinum Neurotoxin target. The last speaker (Prof. Lee) presents the analysis results of antibody repertoires to influenza and SARS-CoV-2 and shows how they can be used in engineering antibodies and vaccines.

South Korea is at the forefront of the customized cosmetic trends. In 2018, Jeju Province and Jeju Technopark launched the national project, financially supported by the Ministry of Trade, Industry and Energy (MOTIE) through the National Innovation Cluster R&D program (P0006712). Next phase research, from 2020 to 2022, is now being conducted to examine the field test and pilot studies of the newly developed products and service platform. This national R&D project was also supported by MOTIE through the same program (P0015296). In this session, recent progress will be presented on the customized cosmetcs funded by the national(MOTIE) project.

Development of Consumables and Raw Materials for Biopharmaceutical Production (바이오소부장 사업 성과발표회 II)

TOC vial, 바이알 스토퍼 : 해당 세부과제는 제품 제작 항목으로써 각각 가격경쟁력 확보 및 국내 수요에 맞는 수급안정성 확보가 중요함. 이에 연구개발과 의약품 제조에 적용할 수 있는 관련 인증 확보 및 GMP시설 인증과 품질 확보가 중요함.

NaOH(수산화나트륨), IPA(이소프로필 알코올): 해당 세부과제는 정제기술 개발 항목으로써 제품의 순도 유지가 중요함. 이에 분순물 제거기술과 정밀분별증류기술에 집중하여 연구함.

Formalin(포르말린): 해당 세부과제는 안정화제로 독성이 큰 메탄올을 대체하여 에탄올을 첨가, 저온에서 Salt가 생기지 않는 조성 연구 등 총 2가지 기술 개발 목표가 있음. 이에 해당 제품의 생산현장 적용가능성을 평가하고 제품에 경쟁성 및 신뢰성 확보를 위한 수요기업 매칭이 중요함

이외 pH, DO 센서 및 비침습적 센서 등 실시간 세포농도 모니터링 기술에 대해 연구개발하여 바이오의약품 원부자재로 사용 범위를 높이고자 함.

And we intend to explore the social role of scientists so that scientists can introduce the latest scientific and technological research trends and their achievements to the public in person, and to make people experience the essential characteristics of science, such as the scientific inquiry process. Experts on science communication who are active in various fields such as SF(Science Fiction), broadcasting and media, writing and lectures are invited as speakers to share experiences on how to create and spread their own scientific and cultural contents. In addition, a variety of methodologies are presented to help make it easier to reach the public. It is intended to provide researchers with insight for scientific communication and to induce participation based on the formation of a consensus on scientific culture activities through this session.

Advanced biotechnology for C1 gas biorefinery

Instruction

Biorefinery process is a promising route for bio-based products from renewable resources but will involve a coordinated effort from pretreatment of biomasses for fermentable sugars, microbial fermentation, and purification of bioproducts in chemical grade. Recently, C1 gas refinery gains much attention for the application of C1 gases including CO and CH4 as promising renewable resources for the production of valuable chemical products in bio, chemical and bio/chemical hybrid processes. In this symposium, we will discuss metabolic engineering strategies for the development of engineered microorganisms to produce chemicals, fuels, and materials from C1 gas resources. As the environmental friendly and carbon closed-loop society has been highlighted, the biomass derived bioplastic and its implementation and expansion are of great importance recently. Corn and sugar cane derived PLAs are representative bioplastic and its scale-up and commercialization are being carried out. However when the bioplastic is discharged into natural environment, physico-chemcial and biological degradation of the plastic are still not extensively studied. Biosystem Development for the Conversion of Unconventional Carbon Sources

Instruction

A carbon-neutral society is a crucial agenda facing humanity today. Amid net zero push, interest in green bioprocesses that allow sustainable development is growing. Innovative strategies for low carbon and energy are required when considering ideal bioprocess. Recently, technology for efficient conversion of gaseous carbon instead of sugar-based conventional carbon sources has been in the limelight, requiring a novel bioreaction system. This session presents the recent progress in bioprocess development for converting carbon dioxide and carbon monoxide, both greenhouse effect-causing gases as well as new carbon sources. Also, an in-depth discussion on the future direction will be held.

Development Status and Prospects of Bioenergy Production Technology

Instruction

Within the carbon-circular economic aspect, bioenergy, a form of renewable energy, has been emphasized due to its carbon neutrality. Interdisciplinary collaborations are needed to produce bioenergy because whole process is put in multiple steps from biomass grow to final products (i.e., transportation fuels, heat, electricity, and chemicals). In this symposium, we will introduce the development of bioenergy research in South Korea during last 30 years. It might be a place to assess the present and future of domestic bioenergy production technology.

Instruction	mRNA vaccines combine desirable immunological properties with an outstanding safety profile and the unmet flexibility of genetic vaccines. Based on in situ protein expression, mRNA vaccines are capable of inducing a balanced immune response comprising both cellular and humoral immunity. In addition, mRNA is an intrinsically safe vector as it is a minimal and only transient carrier of information that does not interact with the genome. In this symposium, we will discuss enabling technologies that have been recently developed to enable mRNA vaccines and related materials.

Instruction	한국생물공학회에서는 한국생명공학연구원 바이오안전성정보센터 및 국가연구안전관리본부의 지원을 받아 유전자변형기술/유전자가위기술 유래 산물의 사업화 촉진을 위해 추진 중인 “바이오위해평가 원팀 리노베이션사업(2021-2023)”을 소개하고 공유하고자 함. 이 사업은 논문·특허 수준에만 머물고 있는 유전자변형기술/유전자가위기술 유래 산물이 위해성평가/심사 장벽을 넘어 사업화까지 갈 수 있도록 바이오원팀(과기정통부, 산업부, 복지부)을 통한 관련 인프라 및 제도개선에 초점을 맞춤. 또한 전 세계적으로 바이오기술의 빠른 발전과 정부의 ‘탄소중립’, ‘화이트바이오산업 활성화 전략’정책 등을 고려해 학계/산업계가 관련 제품의 인·허가에 능동적으로 대응할 수 있도록 가이드 등을 제공하기 위함. 또한 해당 연구개발 및 사업화를 염두에 둔 학계/산업계 관련자들의 현장에서의 애로사항 등을 듣고 해당 사업에 적극적으로 반영 및 개선하고자 하며, 위해성평가/심사, 즉 안전관리에 기반한 사업화 추진은 바이오기술 유래 제품의 국민 안심도 및 수용성 증가, 글로벌 경쟁력 강화에도 기여할 수 있으니 많은 학회 참석자들의 관심 및 참여를 바람.

Instruction	4차 산업혁명에 따라 산업계는 데이터 기반의 제품 및 서비스 개발이 점차 가속화되고 있음. 기초연구 현장에서도 이를 위한 데이터 공유가 활발히 이루어져야 하며, 이를 위하여 연구자들의 인식 제고가 필수적임. 한국생물공학회에서는 과학기술정보통신부 지원을 받아 바이오 데이터 엔지니어 인력양성사업을 수행하고 있음. 사업을 통하여 (1) 청년 예비 바이오 데이터 엔지니어 300명 선발 및 채용, (2) 데이터 수집 및 공유에 대한 인식 제고 활동, (3) 연구 데이터 등록을 추진하고 있으며, 사업 관련 성과발표회를 통하여 지도교수 및 엔지니어들의 사업 활동 내용을 공유하고자 함.

Novel strategies towards enhancing enzymatic properties such as catalytic activity, specificity and stability have opened up a plethora of novel applications for industrial production, agriculture and sustainability. Such improved enzymes are typically identified in high throughput functional screening campaigns by either investigating the vast number of naturally occurring microbial homologs or by exploring the synthetic combinatorial space of given amino acid motifs. Both strategies rely on the ability to generate high numbers of DNA templates. At Twist Biosciences we have revolutionized DNA synthesis in terms of scale, uniformity and error rate. The resulting high-quality products such as clonal genes and DNA variant libraries comprise the ideal basis for high throughput functional screenings for enhanced enzyme variants. In this seminar we will give an introduction to Twist Bioscience, our products and show how we change the game of enzyme engineering.

For the synthetic biologist, biological building blocks are often generated from bacteria. The design-build-test process is utilized in order to engineer these building blocks. This often requires screening and picking colonies of interest. Traditionally, colony picking is performed manually using sterile pipette tips, toothpicks, or inoculation loops, which is a slow, labor-intensive, and time consuming process. Synthetic biology research can, therefore, benefit greatly from the flexibility and throughput of automated colony pickers. Not only will automated colony pickers make the entire process quicker (picking up to 3,000 clones per hour), but the results are more consistent and reliable, increasing the quality of the data produced

기존 이차원 단층세포 배양 기술은, 실험적 편의성과 값싼 비용이라는 장점을 바탕으로 누구나 쉽게 체외에서 특정세포를 배양할 수 있는 연구플랫폼으로 자리잡았으며, 이렇게 배양된 단층배양세포는 개발중인 신약의 성능과 독성을 평가하기위한 보편적인 in vitro 모델로 널리 사용되어 왔습니다. 그러나, 현저하게 낮은 단층배양 세포의 생체 모사성 때문에 이를 기반으로 도출된 연구결과가 실제 임상에서 재현되지 못하는 사례가 빈번하고, 실제로 전임상 및 임상연구의 높은 실패율을 야기하는 치명적인 한계를 보여주고 있습니다. '미니장기' 라고도 불리는 오가노이드 (organoid)는, 줄기세포로부터 형성된 3차원 세포집합체로, 조직을 구성하는 다양한 세포가 실제 조직의 구조적 특성을 재현하고 있기 때문에 기존 단층배양세포의 한계점을 보완하고 조직내의 생리학적 현상을 모사할 수 있는 새로운 형태의 체외배양 모델입니다. 오가노이드는 이러한 우수한 생체모사성으로 인해 환자 맞춤형 치료제와 재생 치료제 개발과 같은 혁신 바이오 기술의 근간이 될 혁신적인 기술로 평가받고 있습니다. ㈜넥스트앤바이오는 오가노이드 배양기술, High Contents Imaging 등 다양한 첨단 융합기술을 바탕으로 환자맞춤형 정밀의약, 정밀진단키트 개발, 오가노이드기반 당뇨치료제 개발 등 미래지향적 의약학 기술개발을 선도하는 것을 목표로 설립된 회사입니다. 본 발표에서는 ㈜넥스트앤바이오가 연구자들께 제공해 드리고 있는 오가노이드 기반 비임상 연구플랫폼과 그로부터 도출된 연구결과를 소개합니다.
